INTRODUCTION
1.1 Background to the Study
Wound infections remain a major clinical and public health challenge worldwide, particularly in developing countries where access to adequate healthcare services and infection control practices is limited. A wound becomes infected when pathogenic microorganisms colonize damaged skin tissue, leading to delayed healing, tissue destruction, and in severe cases, systemic infections such as sepsis. The most commonly implicated pathogens in wound infections include bacteria such as Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, and fungi such as Candida albicans (Bowler et al., 2001).
The increasing prevalence of antimicrobial-resistant pathogens has complicated the management of wound infections. Antibiotic-resistant bacteria, including Methicillin-Resistant Staphylococcus aureus (MRSA) and multidrug-resistant Pseudomonas aeruginosa, have become a major concern in healthcare settings due to their ability to resist conventional treatment options. Similarly, opportunistic fungal infections have also increased, particularly among immunocompromised patients (WHO, 2023). This dual burden of bacterial and fungal resistance has made wound management more difficult and expensive.
Conventional antibiotics and antifungal drugs are widely used in treating wound infections; however, their effectiveness is increasingly being compromised by resistance development, misuse, and overprescription. In addition, many synthetic antimicrobial agents are associated with adverse side effects and high cost, making them less accessible in low-resource settings. These challenges have created an urgent need for alternative, safe, and effective antimicrobial agents.
Medicinal plants have gained significant attention as potential sources of new antimicrobial compounds. Plants produce a wide variety of secondary metabolites known as phytochemicals, which include flavonoids, tannins, saponins, alkaloids, phenols, and terpenoids. These compounds have been shown to possess antimicrobial, antioxidant, anti-inflammatory, and wound-healing properties (Cowan, 1999). The use of plant-based remedies in traditional medicine for treating wounds and infections has been practiced for centuries across different cultures.
One of the most widely studied medicinal plants is Aloe vera (Aloe barbadensis Miller), a succulent plant belonging to the Liliaceae family. Aloe vera gel is extracted from the inner leaf pulp and is widely known for its therapeutic properties, including wound healing, anti-inflammatory, antimicrobial, and skin-soothing effects. The gel contains bioactive compounds such as acemannan, anthraquinones, vitamins, enzymes, amino acids, and polysaccharides that contribute to its medicinal value (Surjushe et al., 2008).
Aloe vera has been traditionally used for treating burns, cuts, ulcers, and various skin infections. Its wound healing ability is attributed to its capacity to promote collagen synthesis, enhance tissue regeneration, and improve moisture retention at the wound site. In addition, Aloe vera exhibits antimicrobial properties that help prevent infection and support faster wound healing (Chandegara & Varshney, 2013).
Scientific studies have reported that Aloe vera gel possesses both antibacterial and antifungal activities against a range of pathogenic microorganisms. It has been shown to inhibit the growth of bacteria such as Staphylococcus aureus and Escherichia coli, as well as fungi such as Candida albicans. These effects are believed to be due to the synergistic action of its phytochemical constituents (Hamman, 2008).
Despite its widespread use, variations in extraction methods, concentrations, and microbial strains have led to inconsistent findings regarding the antimicrobial effectiveness of Aloe vera gel. Some studies report strong antimicrobial activity, while others suggest moderate or weak effects, highlighting the need for further investigation under controlled laboratory conditions.
The evaluation of both antibacterial and antifungal potentials of Aloe vera gel is particularly important because wound infections are often polymicrobial in nature, involving both bacterial and fungal pathogens. Therefore, a broad-spectrum antimicrobial agent would be highly beneficial in wound management.
In developing countries, Aloe vera is readily available, affordable, and widely used in traditional medicine. This makes it a promising candidate for developing alternative antimicrobial therapies, especially in resource-limited healthcare systems where access to modern drugs may be restricted.
Given the rising challenge of antimicrobial resistance and the need for effective wound management strategies, there is a strong justification for exploring plant-based alternatives. This study therefore focuses on the evaluation of the antifungal and antibacterial potentials of Aloe vera gel extract on pathogenic microorganisms associated with wound infections.
1.2 Statement of the Problem
Wound infections continue to be a significant healthcare problem globally, contributing to prolonged hospital stays, increased healthcare costs, and high morbidity rates. The situation is further complicated by the rising prevalence of antibiotic-resistant and antifungal-resistant microorganisms. Pathogens such as MRSA, multidrug-resistant Pseudomonas aeruginosa, and Candida albicans have shown increasing resistance to conventional antimicrobial agents, making treatment increasingly difficult (WHO, 2023).
The overuse and misuse of antibiotics and antifungal drugs have accelerated the development of resistance among pathogenic microorganisms. As a result, standard treatments are becoming less effective, leading to treatment failures and chronic wound conditions. This has created an urgent need for alternative therapeutic options that are both effective and accessible.
Although synthetic antimicrobial agents are widely used in clinical practice, they are often associated with side effects such as allergic reactions, toxicity, and high cost. In low-income regions, limited access to these drugs further worsens the burden of wound infections. These challenges highlight the need for safe, affordable, and effective natural alternatives.
Aloe vera gel has been widely used in traditional medicine for the treatment of wounds and skin infections. It is believed to possess both antibacterial and antifungal properties, making it a potential broad-spectrum antimicrobial agent. However, despite its widespread use, there is still insufficient scientific evidence confirming its effectiveness against clinically relevant wound pathogens under standardized conditions.

Some researchers report strong inhibitory effects, while others observe weak or negligible activity. These inconsistencies may be due to differences in extraction methods, concentration levels, or microbial strains used in previous studies.
Another gap in knowledge is the limited research focusing on the combined evaluation of both antifungal and antibacterial properties of Aloe vera gel against wound-associated pathogens. Most studies tend to focus on either bacteria or fungi, rather than both, despite the polymicrobial nature of wound infections.
Therefore, this study seeks to address these gaps by evaluating the antifungal and antibacterial potentials of Aloe vera gel extract on pathogenic microorganisms associated with wound infections, with the aim of providing scientific evidence to support or refute its traditional medicinal use.
1.3 Aim of the Study
The aim of this study is to evaluate the antifungal and antibacterial potentials of Aloe vera gel extract on pathogenic microorganisms associated with wound infections.
1.4 Objectives of the Study
The specific objectives are to:
- Determine the antibacterial activity of Aloe vera gel extract against bacterial pathogens associated with wound infections.
- Determine the antifungal activity of Aloe vera gel extract against fungal pathogens associated with wound infections.
- Evaluate the extent of inhibition produced by Aloe vera gel extract on selected microorganisms.
- Assess the potential of Aloe vera gel as a broad-spectrum antimicrobial agent for wound management.

1.6 Significance of the Study
This study will contribute to scientific knowledge on the antimicrobial properties of Aloe vera gel and its potential application in wound management. It will be beneficial to microbiologists, pharmacologists, healthcare practitioners, and pharmaceutical industries in the development of alternative antimicrobial agents.
The findings may also support the use of Aloe vera in developing affordable, plant-based wound care products, especially in resource-limited settings where access to modern antimicrobial drugs is restricted.
1.7 Scope of the Study
This study is limited to the evaluation of the antibacterial and antifungal activities of Aloe vera gel extract against selected pathogenic microorganisms associated with wound infections. It does not include in vivo or clinical trials.
